Abstract: This article describes the analysis of the chemical profile of a phytotherapeutic supplement of Ilex paraguariensis that was rapidly conducted by using the LC-SPE/NMR hyphenated technique providing the rapid separation and identification of eight organic compounds in a single experiment. Therefore, bench isolation methods were not necessary. The high selectivity achieved in the chromatographic separation combined with adequate retention by SPE and the application of cryogenic technology of high magnetic field NMR (14.1 Tesla) allowed to reach conclusive data on the structure of the substances found, thus the use of mass spectrometry was unnecessary.
