Abstract: In the viticulture, the micropropagation techniques become indispensable for the obtaining in large scale material of good quality fitossanitary. The present work had as objective to study the effect of adenine sulphate, associated to BAP on the multiplication in vitro of two grapevine rootstock. Nodal segments of grapevine rootstock ‘VR043-43’ (Vitis vinifera x V. rotundifolia) and ‘R110’ (Vitis berlandieri x V. rupestris), with 2 cm length, originating from buds established in vitro were excisaded and introduced in tubes containing 15 mL of DSD1 culture medium. The experiment was planed to study two rootstocks effect when different concentrations of adenine sulphate (0; 20; 40; 60 and 80 mg L-1) and concentrations of BAP (0; 0.5 and 1.0 mg L-1), in all possible combinations, added to the culture medium. The medium cultures were added of 20 g L -1 of sucrose, solidified with 6 g L-1 of agar and the adjusted pH for 6.4, before the sterilization to 121ºC and 1 atm for 20 minutes. After the inoculation, the tubes were transferred for growth room to 25±2ºC, irradiance of 35 μmol.m-2.s-2 and photoperiod of 16 hours daily, staying in these conditions per 70 days. The experiment was arranged in a completely randomized design, being used four repetitions with twelve buds per treatment. Data on number of leaves, length of the aerial part, weigh of the fresh matter of the aerial part and callus, were recorded. Better results in the micropropagation of ‘VR043-43’’ and ‘R110’ were obtained in the culture medium DSD1 without the addition of adenine sulphate and with 1.0 mg L-1 of BAP.
