Abstract: This study aimed at verifying the ploidy level of diploid, mixoploid and tetraploid banana plants (Musa acuminata Colla) obtained four years ago and its relationship with the leaf anatomy. Banana plantings were submitted to chromosome duplication in in vitro conditions by exposure of the apices to colchicine (concentrations and varied times). After the process, diploids, mixoploids and tetraploids were counted. After four years, the ploidy of the materials was again verified by flow cytometry and the anatomy of the leaves was performed to determine a possible relationship. Stomatal and cytometric analysis proved compatible for ploidy verification of tetraploid plants. The original tetraploid plants and most of the mixoploids remained genetically stable until the end of this study. However, the shoots were diploid and mixoploids.
