Resumo: Studies on structural changes resulting from in vitro culture procedures allow us to understand its effects, develop more efficient protocols and to optimize the micropropagation techniques. Thus the objective was to evaluate the influence of the seal and substrate employed during the pre-acclimatization on stem anatomy of ipê-amarelo plants. Plants previously established in vitro were inoculated in test tubes with cotton seal, cover free and with parafilm and substrates with agar, vermiculite and Plantmax®. After 45 days of pre-acclimatization, internodal segments of plants of each treatment were extracted for construction of stem cross sections and for comparison the same procedure was carried out in plants in vivo. The experiment was conducted in completely randomized 3x3 factorial scheme (three types of sealing and of substrate) with additional treatment (in vivo). The use of cotton or cover without parafilm combined with agar during the pre-acclimatization led to stem modifications which favored the acclimatization of ipê-amarelo.
