Resumo: | The blackberry micropropagation is used, mainly to produce virus-free plants in a short period of time. With the objective to improve the techniques of in vitro blackberry cv. Tupy cultivation, different concentrations of sucrose (0, 15, 30, 45 and 60 g L-1) and grape juice (SuFresh ) (0, 5, 10, 15 and 20%) added to MS medium supplemented with 1.0 mg L-1 BAP, 6.0 g L-1 agar and pH adjusted to 5.8 prior sterilization at 121ºC and 1 atm for 20 minutes were tested. Nodal segments, originated from in vitro plants were excised and inoculated in tubes containing 15 mL of culture medium and then transferred to a growth room with temperature of 25 ± 2ºC, irradiance of 32 mol m-2 s-1 and photoperiod of 16 hours. The experiment was a complete randomized design with four replicates and 12 segments each. The experiment was evaluated after 70 days of in vitro cultivation. Best results for micropropagation of the blackberry cv. Tupy were obtained using high (45-60 g L-1) sucrose concentrations associated with reduced (0-5%) concentrations of grape juice. |
