Abstract: In vitro culture has been applied in many cultures in order to obtain matrix plants, especially in ornamentals. The objective of this study was to determine the best concentration of MS medium for the propagation of seedlings of chrysanthemum in two cultivation environments (growth room and greenhouse). Nodal segments of chrysanthemum plantlets were inoculated into flasks containing 0, 25, 50, 75 and 100% MS medium. The experiment was conducted in DIC 5 x 2, resulting in 10 treatments and 7 replicates with 5 nodal segments. After 45 days, the following characteristics: number of leaves, shoots and roots and shoot length were evaluated. The results showed superiority of the regime of natural light in all treatments with an increase of 96.58% in the number of leaves, 80% for shoots and roots, and 12% for shoot length when compared with plants incubated under artificial light.
