Resumo: The Serra da Boa Esperança State Park (PESBE) was created in 2007 in an ecotonal area between Cerrado and Atlantic Forest, two biodiversity hotspots for global conservation. Located in the southern region of the Minas Gerais State and by side to Furnas hydroelectric plant, Serra da Boa Esperança is considered a Natural Heritage Site. Many studies are carrying out in the park, but this is the first on medium and large mammals, which will bring better knowledge about species in wildlife protected areas in Minas Gerais, helping to fill in gaps about the richness of ecotonal regions. This study aimed to inventory the medium and large mammals of PESBE and part of its surroundings. Collection methods included camera traps, searching for traces, sightings, and employee reports to 29 locations sampled. The collector and richness estimated curves were performed. The estimated richness index used was the 1st order Jackknife. A total of 16 native and three exotic species were registered, representing 71% of the estimated richness, including exotic mammals. The mammals composition revealed relevant and endangered species like mesopredators and top-of-the-chain predators. The low amount of wildlife-protected areas and other natural areas in connectivity with PESBE and the high fragmentation make the site a refuge for mammals.
