Pleurotus spp. and their proteases: a new approach to alternative helminth control in agriculture and livestock farming

dc.description.abstractThe use of edible mushrooms and their cell-free proteolytic crude extract, especially from species of the genus Pleurotus, as biological and biochemical control agents presents itself as a sustainable alternative for parasite management in agriculture and livestock. In this context, the general objective of this thesis was to optimize enzyme production by Pleurotus spp. and evaluate both the antiparasitic activity of the fungi and their proteolytic enzymes on eggs and juveniles of plant-parasitic nematodes and larvae of gastrointestinal parasites of animals. The first article presented a review of the proteases produced by Pleurotus spp., addressing their production, biochemical properties, and biotechnological applications. The second article investigated the nematophagous activity of several species of the genus Pleurotus and the nematicidal activity of their proteases against Panagrellus sp. Significant correlations were observed between both assays. The third article evaluated the in vitro nematophagous activity of P. djamor and the nematicidal activity of its proteases on infective larvae of Trichostrongylus spp. and Strongyloides papillosus, showing a 75% reduction of nematodes in the nematophagous assay and 57% in the nematicidal assay. In the fourth article, the optimization of protease production by P. djamor was carried out using solid-state fermentation with agricultural residues, specifically wheat bran. The results indicated that moisture levels significantly influenced enzyme production. Also in this article, the proteases demonstrated significant in vitro anthelmintic activity against eggs of T. solium and Moniezia sp., with reductions of 33.44% and 45.43%, respectively. The fifth article analyzed the proteolytic profile and the nematicidal action of P. djamor proteases against larvae of Haemonchus spp. and Trichostrongylus spp., resulting in a 35% reduction in larvae. Zymogram analysis revealed at least two proteases with molecular weights between 75 and 100 kDa. The sixth article evaluated the nematicidal and ovicidal activity of P. djamor proteases on Meloidogyne incognita in vitro and under greenhouse conditions, demonstrating a 100% reduction in juveniles and 22% in eggs in vitro. Both extracts containing active proteases and denatured proteases of P. djamor significantly reduced egg numbers by 94% and 97%, respectively, and decreased gall formation under greenhouse conditions. Finally, the seventh article investigates the combined use of P. djamor proteases and papain, aiming at the development of more effective formulations for nematode control. The results of the articles highlight the potential of the fungus Pleurotus spp. and its proteases as a sustainable alternative for the management of important parasites present in agriculture and livestock.

dc.description.resumoO uso de cogumelos comestíveis e de seu extrato bruto proteolítico livre células, especialmente de espécies do gênero Pleurotus, como agentes de controle biológico e bioquímico, apresenta-se como uma alternativa sustentável para o manejo de parasitas na agropecuária. Nesse sentido, essa tese teve como objetivo geral otimizar a produção de enzimas por Pleurotus spp. e avaliar tanto a atividade antiparasitária dos fungos quanto de suas enzimas proteolíticas sobre ovos e juvenis de nematoides parasitas de plantas e larvas de parasitas gastrointestinais de animais. O primeiro artigo apresentou uma revisão das proteases produzidas por Pleurotus spp., abordando sua produção, propriedades bioquímicas e aplicações biotecnológicas. O segundo artigo investigou a atividade nematófaga de várias espécies do gênero Pleurotus e nematicida de suas proteases contra Panagrellus sp. Correlações significativas foram observadas entre ambos ensaios. O terceiro artigo avaliou a atividade nematófaga in vitro de P. djamor e nematicida de suas proteases sobre larvas infectantes de Trichostrongylus spp. e Strongyloides papillosus, evidenciando uma redução de 75% dos nematoides no ensaio nematófago e 57% no ensaio nematicida. No quarto artigo, foi realizada a otimização da produção de proteases por P. djamor usando fermentação em estado sólido com resíduos agrícolas, especificamente farelo de trigo. Os resultados indicaram que os níveis de umidade influenciaram significativamente a produção de enzimas. Ainda neste artigo, as proteases demonstraram atividade anti-helmíntica in vitro significativa contra ovos de T. solium e Moniezia sp., com reduções de 33,44% e 45,43%, respectivamente. O quinto artigo analisou o perfil proteolítico e a ação nematicida das proteases de P. djamor contra larvas de Haemonchus spp. e Trichostrongylus spp., resultando em uma redução significativa de 35% das larvas. A análise por zimograma revelou pelo menos duas proteases com pesos moleculares entre 75 e 100 kDa. O sexto artigo avaliou a atividade nematicida e ovicida das proteases de P. djamor sobre Meloidogyne incognita in vitro e em condições de estufa, demonstrando uma redução de 100% em juvenis e de 22% em ovos in vitro. Tanto os extratos com proteases ativas quanto com proteases desnaturadas de P. djamor reduziram significativamente o número de ovos em 94% e 97%, respectivamente, além de diminuírem a formação de galhas em condições de estufa. Por fim, o sétimo artigo investiga o uso combinado de proteases do P. djamor e da papaína, visando o desenvolvimento de formulações mais eficazes para o controle de nematoides. Os resultados dos artigos evidenciam o potencial do fungo Pleurotus spp. e de suas proteases como alternativa sustentável para o manejo de parasitas importantes presentes agropecuária.
