dc.description.abstractThe ex situ preservation and rational use of Anadenanthera colubrine demand conclusive information related to seed storage. This study aimed determines a protocol for cryopreserving embryonic axes of A. colubrine seeds. Embryonic axes were treated with three concentrations (5, 10, and 15%) of DMSO and glycerol and maintained in liquid nitrogen (-196°C), ultra freezer (-80°C) and freezer (-20°C) for 15, 30, and 150 days. Moisture content of the embryonic axes was determined before and after each storage period. Embryonic axes immersed in chemical cryoprotectants and maintained at -196, -80, and -20°C, absorbed the solutions and had moisture contents higher than 65%. Under these conditions, no germination was observed. Embryonic axes that were stored in the absence of cryoprotectants presented viability around 100%, independently of the cryostorage period.pt_BR
