Resumo: Fish are sensitive animals and susceptible to stress during handling process. The use of anesthesia is routine in fish farming sectors, such as 2- phenoxyethanol for facilitating the process and minimize the effects on the physiological and behavioral aspects in fish. However, chemicals used during the production of fish may accumulate in the organism of the animals. According to the World Health Organization (WHO), no food for human consumption must contain residues of products used during their production. To that end, this work was carried out residual quantification of 2- phenoxyethanol anesthetic in fish fillets of Nile tilapia species (Oreochromis niloticus), anesthesia concentrations being analyzed between 450 and 1050 µg L -1 and residual periods 0 - 24 h. For the preparation of samples microextraction techniques were employed in solid phase (SPME) and single drop (SDME), both in headspace with separation and detection by chromatography coupled gas phase will mass spectrometry (GC/MS). For the development of analytical methodologies, three parameters (salt content, temperature and extraction time) were evaluated for both multivariate way to have influence on the extraction efficiency of 2-phenoxyethanol. In planning the analysis was employing the central compound design (CCD), which made it possible to analyze the variables jointly and strategically, resulting in accurate measurements with less analysis. The developed methods were validated and applied to the residual quantification of 2-phenoxyethanol in Tilapia fillets, with considerable precision and accuracy.
