Abstract: The use of molecular markers variation has a long tradition in population genetics, and more recent application in forest tree improvement, management and conservation genetics. It is know that the isozymes are reliable markers for population genetics studies, species and hybrids identification, because of the rapidity and low economic cost of electrophoretic methods. The aim of this study was to investigate and to select extraction protocols and enzymatic systems of several individuals of the tree Protium spruceanum for genetic diversity studies. Extraction protocols and 14 enzymatic systems were analyzed by using the starch gel electrophoresis technique on plant leaves. Among all extraction protocols and enzymatic systems investigated here, the buffer systems n° 1 of Alfenas and ADH, GDH, GLDH, GTDH, MDH, PER, SDH e SKDH enzymatic systems were the most suitable for identifying P. spruceanum isozymes. The eight enzyme systems selected showed ten loci that could be interpreted and 20 alleles.
