Abstract: This research was carried out to test sucrose concentrations and types of sealing bottles used for in vitro cultivation of teak (Tectona grandis L.f.). The experimental design was completely randomized in a 2 x 6 factorial, two types of seal (lid with filter and lid without filter) and six sucrose concentrations (0, 6, 12, 18, 24 and 30 mg.L-1 ) totaling twelve treatments with ten repetitions each. The explants were cultured on MS medium and kept for 35 days in a growth chamber with a temperature of 30 ± 2 ºC and 16 h photoperiod. The following variables were determined: length of shoot (cm), number of internodes and dry weight of shoots (mg). The best results were obtained with the use of the cover with filter. For each variable, the best sucrose concentration was different. At the concentration of 17 mg.L-1 sucrose, sprouts showed 4.1 cm in length. At the concentration of 18 mg.L-1 , the number of internodes, was 4.6 and for the dry matter, the best concentration was 30 mg.L-1 , with an average of 0.0162 mg. The best results were obtained with 18 mg.L-1 sucrose and the plastic cover with filter.
