Abstract

Syngonanthus paepalophyllus is an endemic species from Brazil exploited for ornamental purposes, which has put this species under serious extinction risks. Micropropagation has been performed for commercial production and preservation of these plants, but they have been shown to be sensitive to the salt concentration, thus, adjustment is necessary. Although, there are few studies on micropropagation of plantlets of the Syngonanthus genus, there is no study on in vitro growth of S. paepalophyllus. This study aimed to evaluate the different culture media for in vitro seeds germination and establishment of plantlets of S. paepalophyllus. To study the in vitro seed germination 4 lots with 50 seeds were used. The seeds were placed on WPM culture medium and a control consisting of only deionized water. During the 30-day period, the number of germinated seeds was daily recorded. To evaluate the S. paepalophyllus in vitro development MS and WPM with 50 and 100% of salt concentrations were tested. After 30 days of inoculation, the number of leaves, chlorotic leaves, length of plantlets, length of roots, fresh and dry mass weight of the plantlets was evaluated. The maximum percentage of germination was 70.9 and 60.5% on media constituded of water and WPM, respectively. The number of leaves was 16.3 and 9.5 leaves on WPM and MS, respectively. Pronounced chlorosis was observed on leaves grown on complete MS medium. The cultivation on 50% WPM propitiated shoot length, higher plant and root lengths in S. paepalophyllus. However, the complete WPM allowed the highest fresh mass production.
